Ocus切片扫描用于AUB异常子宫出血疾病的相关研究


AUB(Abnormal Uterine Bleeding)异常子宫出血(AUB)是妇科常见的症状和体征。该疾病分型较多,如AUB-A型(子宫腺肌病所致)、AUB-L型(子宫平滑肌瘤所致)、AUB-P型(子宫内膜息肉所致)及AUB-E型(病因多重且易忽略)等。现阶段,临床的治疗手段多为子宫内膜切除或子宫整体切除。
 

 
2021年3月发表在《Reproductive Biology and Endocrinology》(生殖生物学和内分泌学)的这篇文章,来自浙江大学医学院附属妇产科医学院(暨浙江省妇女保健院)周坚红研究团队。对于AUB-E型疾病,该团队结合多年的临床诊断和研究,利用无标记的蛋白质组学揭示SMC1A(染色体修复蛋白1A亚型)在AUB-E型体内表达下调。这一结论同时得到Western Blot和免疫组化两种方法的一致性验证。

文章中包含了详细的的实验流程如图 1,对AUB-E病患(实验组)和健康女性(对照组)的子宫内膜增生组织采样,HPLC高效液相色谱解析并分离出样品内的蛋白。这些蛋白通过Liner Ion Trap(LTQ)-Orbitrap 离子阱和高场Orbitrap组合式质谱的检测(如定性/定量),并完成相应数据的比对和处理。基于以上数据,Cytoscape软件(对蛋白质相互作用路径的预测分析)的富集分析法得DEPs(差异表达蛋白)结果,同SMC1A蛋白相关(具体图示见原文)。随后,大量的生信分析得出结论:SMC1A(染色体修复蛋白1A亚型)在AUB-E型体内表达下调,并得到Western blot和免疫组化两种方法的双重验证。其中,Ocus(芬兰Grundium)用于对免疫组化(IHC方法)的组织切片,进行数字化扫描和图像保存、导出,如图2。图像分析得到AUB-E型病患的SMC1A 表达下调,且对比Stromal间质细胞,Glandular epithelium(腺上皮细胞) 的下调更明显(促进了大量腺上皮细胞增殖)。
